A role for histone-like protein H1 (H-NS) in the regulation of hemolysin expression by Serratia marcescens BJMBR
Franzon,J.H.; Santos,D.S..
The histone-like protein H1 (H-NS) is an abundant structural component of the bacterial nucleoid and influences many cellular processes including recombination, transcription and transposition. Mutations in the hns gene encoding H-NS are highly pleiotropic, affecting the expression of many unrelated genes. We have studied the role of H-NS on the regulation of hemolysin gene expression in Serratia marcescens. The Escherichia coli hns mutant carrying S. marcescens hemolysin genes on a plasmid constructed by ligation of the 3.2-kb HindIII-SacI fragment of pR02 into pBluescriptIIKS, showed a high level of expression of this hemolytic factor. To determine the osmoregulation of wild-type and hns defective mutants the cells were grown to mid-logarithmic phase in...

Biological activity of Serratia marcescens cytotoxin BJMBR
Carbonell,G.V.; Amorim,C.R.N.; Furumura,M.T.; Darini,A.L.C.; Fonseca,B.A.L.; Yano,T..
Serratia marcescens cytotoxin was purified to homogeneity by ion-exchange chromatography on a DEAE Sepharose Fast Flow column, followed by gel filtration chromatography on a Sephadex G100 column. The molecular mass of the cytotoxin was estimated to be about 50 kDa. Some biological properties of the cytotoxin were analyzed and compared with well-characterized toxins, such as VT1, VT2 and CNF from Escherichia coli and hemolysin produced by S. marcescens. The sensitivity of the cell lines CHO, HeLa, HEp-2, Vero, BHK-21, MA 104 and J774 to the cytotoxin was determined by the cell viability assay using neutral red. CHO and HEp-2 were highly sensitive, with massive cellular death after 1 h of treatment, followed by BHK-21, HeLa, Vero and J774 cells, while MA 104...

Characterization and comparison of Serratia marcescens isolated from edible cactus and from silkworm for virulence potential and chitosan susceptibility BJM
Li,Bin; Yu,Rongrong; Liu,Baoping; Tang,Qiaomei; Zhang,Guoqing; Wang,Yanli; Xie,Guanlin; Sun,Guochang.
Representative strains of Serratia marcescens from an edible cactus plant and silkworms were characterized and a comparison based on their cellular fatty acid composition, 16S rRNA and groE gene sequence analysis as well as silkworm virulence and chitosan susceptibility was carried out. Results from this study indicate that there are no significant differences between the phenotypic and molecular characterization, virulence and chitosan susceptibility of the S. marcescens strains from the cactus plant and silkworms. Silkworms inoculated with S. marcescens from either plant or silkworm resulted in nearly 100% mortality. Chitosan solution exhibited strong antibacterial activity against S. marcescens. This activity increased with the increase of chitosan...

Detection of cytotoxic activity on Vero cells in clinical isolates of Serratia marcescens BJMBR
Carbonell,G.V.; Alfieri,A.F.; Alfieri,A.A.; Vidotto,M.C.; Levy,C.E.; Darini,A.L.C.; Yanaguita,R.M..
Cytotoxin production was studied in 60 Serratia marcescens strains isolated from hospitalized patients. Association of cytotoxic activity with serotype, source of isolation and presence of plasmids was also evaluated. Thirteen of the 60 S. marcescens strains produced a cytotoxic effect on Vero cells. These strains were isolated from distinct clinical sources and classified into seven different serotypes (O1:H7; O4:NM; O10:NT; O19:NM; O6,14:H4; O6,14:NM and O6,14:H1). No relationship was observed between cytotoxic activity and clinical source or serotypes of the strains. Plasmids from five cytotoxin-producing S. marcescens strains were transferred to E. coli K12/711. The transconjugants did not exhibit cytotoxicity, indicating that the cytotoxic effect is...

Draft genome sequence of a GES-5-producing Serratia marcescens isolated in southern Brazil BJM
Nodari,Carolina Silva; Siebert,Marina; Matte,Ursula da Silveira; Barth,Afonso Luís.
Abstract Serratia marcescens is a Gram-negative rod intrinsically resistant to polymyxins and usually associated with wound, respiratory and urinary tract infections. The whole genome of the first GES-5-producing S. marcescens isolated from a Brazilian patient was sequenced using Ion Torrent PGM System. Besides blaGES-5, we were able to identify genes encoding for other β-lactamases, for aminoglycoside modifying enzymes and for an efflux pump to tetracyclines.

Enhanced production of prodigiosin by Serratia marcescens MO-1 using ram horn peptone BJM
Kurbanoglu,Esabi Basaran; Ozdal,Murat; Ozdal,Ozlem Gur; Algur,Omer Faruk.
This work addresses the production of prodigiosin from ram horn peptone (RHP) using MO-1, a local isolate in submerged culture. First, a novel gram-negative and rod-shaped bacterial strain, MO-1, was isolated from the body of the grasshopper (Poecilemon tauricola Ramme 1951), which was collected from pesticide-contaminated fields. Sequence analysis of 16S rDNA classified the microbe as Serratia marcescens. The substrate utilization potential (BIOLOG) and fatty acid methyl ester profile (FAME) of S. marcescens were also determined. The effect of RHP on the production of prodigiosin by S. marcescens MO-1 was investigated, and the results showed that RHP supplementation promoted the growth of MO-1 and increased the production of prodigiosin. A concentration...

Highly Solvent Tolerance in Serratia marcescens IBBPo15 BABT
Stancu,Mihaela Marilena.
ABSTRACT The aim of this study was to investigate the solvent tolerance mechanisms in Serratia marcescens strain IBBPo15 (KT315653). Serratia marcescens IBBPo15 exhibited remarkable solvent-tolerance, being able to survive in the presence of high concentrations (above 40%) of toxic organic solvents, such as cyclohexane, n-hexane, n-decane, toluene, styrene, and ethylbenzene. S. marcescens IBBPo15 produced extracellular protease and the enzyme production decreased in cells exposed to 5% cyclohexane, n-hexane, toluene, styrene, and ethylbenzene, as compared with the control and n-decane exposed cells. S. marcescens IBBPo15 cells produced carotenoid pigments and alteration of pigments profile (i.e., phytoene, lycopene) were observed in cells exposed to 5%...

Spondylodiscitis associated with recurrent Serratia bacteremia due to a transjugular intrahepatic portosystemic shunt (TIPS): a case report BJID
Marques,Nuno; Sá,Rosa; Coelho,Filomena; Cunha,Saraiva da; Meliço-Silvestre,A..
We report a case of spondylodiscitis caused by multiresistant Serratia marcescens in a cirrhotic patient who had several Serratia bacteremias after the placement of a transjugular intrahepatic portosystemic shunt (TIPS) device. We concluded that an endovascular stent that can not be removed makes management of recurrent bacteremia difficult. Furthermore, back pain due to bacteremia is indicative of spondylodiscitis. Serratia marcescens can be an aggressive pathogen, causing spinal infection.
